The scene
Titan, in Infinity War, where two teams who have never met land on the same planet and assume the worst. Star-Lord carries twelve of the forty-three lines, Iron Man eight, Spider-Man and Drax seven each, Doctor Strange six, Mantis three. The first minute is an ambush: Drax yelling as he leaps, Spider-Man begging nobody to put eggs in him, a taser noise, Star-Lord telling everyone to calm down. Then the standoff, where everyone has a weapon on someone and nobody knows who anyone is.
It's an ensemble comedy hiding inside an action film. The Gamora exchange stacks three lines, Star-Lord's question, Iron Man's correction, Drax topping both with Why is Gamora?
, and that triplet is the rhythm of the whole scene: someone says a thing, someone reframes it, someone makes it dumber. Star-Lord's insistence that he's from Missouri rather than Earth, Iron Man's contempt, and Mantis and Strange being the only two people trying to lower the temperature all fit inside two minutes fifteen.

Performing it
The Gamora triplet is the test. Three voices, three beats, and the third has to be delivered with total sincerity or it dies; Drax never knows he's funny. Star-Lord's threat to French fry Spider-Man is his one long, fast line, and his Missouri answer wants wounded pride. Spider-Man's lines are mostly panic, including one that's literally a taser sound. Drax's yells during the ambush are big and short, so keep the mic back for those. Six voices over two players means everyone doubles; putting Drax and Mantis in one mouth and Star-Lord and Iron Man in the other keeps the standoff bouncing between you.
