The scene
Waternoose confesses on camera and finds out the room isn't real. Eighteen of the thirty-four lines are his, Sulley has eight, Mike Wazowski two, and a handful go to Boo and the agents. Eighty seconds.
Sulley pushes him, and Waternoose says the thing the whole film has been waiting for: he'll kidnap a thousand children before he lets the company die, and silence anyone in his way. Then the simulation terminates, Mike announces he spotted several big mistakes, and the confession is played back to him three times. He is arrested telling Sulley he destroyed the company and asking where everyone will get their scream now.

Performing it
Waternoose has to be genuinely frightening for the first half — this is a grandfather figure admitting to a crime, and the more reasonable it sounds the worse it is. The confession line is delivered three more times as a recording, and the fun is deciding how identical to make those; matching your own delivery exactly is the funnier choice. Mike's two lines are the turn and should be as light and pleased as possible, a man enjoying his work. The final speech is bluster from someone who has already lost, so let it get shriller rather than louder.
